MT-2 Overview

MT-2 (Melanotan II) is a synthetic cyclic peptide developed as an analog of alpha-melanocyte-stimulating hormone (α-MSH), an endogenous peptide belonging to the melanocortin family. Through structural modifications designed to improve stability and receptor affinity, MT-2 has become a widely studied research peptide in investigations involving melanocortin receptor biology. Research involving MT-2 spans pigmentation biology, neuroendocrinology, metabolism, and receptor pharmacology. Experimental studies continue to evaluate its interactions with melanocortin receptor subtypes and the downstream signaling pathways involved in pigmentation, appetite regulation, and other physiological processes.

History

Melanotan II was developed during research aimed at creating more stable analogs of naturally occurring α-MSH. Scientists modified the native peptide to increase resistance to enzymatic degradation while enhancing receptor activity, resulting in the cyclic peptide now known as Melanotan II. Since its development, MT-2 has become an important research tool for studying melanocortin receptor function and has been investigated across numerous experimental models involving pigmentation, neurobiology, metabolism, and endocrine signaling.

Research Findings

MT-2 is widely investigated for its interactions with melanocortin receptor signaling pathways, particularly MC1R, MC3R, MC4R, and MC5R. Experimental studies suggest these receptors participate in numerous physiological processes involving pigmentation, energy homeostasis, inflammatory regulation, and neuroendocrine function. Current evidence consists primarily of biochemical, cellular, animal, and early clinical investigations. Ongoing research continues to explore receptor selectivity, downstream signaling mechanisms, and the broader physiological roles of melanocortin receptor activation.
